    Antimicrobial resistance is a growing health problem worldwide. The World Health Organization has included among its most critical priorities the development of new antibacterial agents for the treatment of Enterobacteriaceae resistant to carbapenems (CRE), as this type of resistance has minimized the usefulness of many commercially available drugs, limiting the therapeutic options available.

    The main mechanism of resistance to carbapenems is based on the production of the carbapenemase enzyme, that is able to hydrolyze carbapenems and other β-lactam antibiotics. The encoding genes for this enzyme are associated with plasmids and therefore horizontally transferable and they are included in three of the four classes of β-lactamases (A, B and D), according to the Ambler classification.

    KPC class A enzymes are currently the predominant carbapenemases reported worldwide among Enterobacteriaceae. VIM, IMP and NDM are the most geographically widespread metallo-β-lactamases. As for class D carbapenemases, OXA-48-like carbapenemases are the most common among Enterobacteriaceae.

    Together with CRE, Acinetobacter baumannii is considered as one of the main hospital-acquired pathogens capable of resisting the main antimicrobials and surviving in the healthcare environment. OXA-type carbapenemases constitute the most widespread mechanism of resistance to carbapenems in this species.

    Another resistance that has emerged in recent years is colistin resistance, which is considered a problem due to the lack of alternative antibiotics. Some bacteria, including Pseudomonas aeruginosaAcinetobacter baumannii, and members of Enterobacteriaceae, such as E. coliSalmonella spp. and Klebsiella spp. have acquired resistance to colistin. Colistin is an effective antibiotic for the treatment of most multidrug-resistant Gram-negative bacteria. Horizontal transfer of colistin resistance is given by mcr genes.

    The Candida genus includes a variety of microorganisms, normally commensal of human mucous membranes. More than 200 species are recognized, however, only 10% are classified as pathogenic to humans. Microorganisms of the Candida genus can cause superficial infections, which can involve the mucous membranes, the skin and in the most serious cases also invasive systemic infections characterized by high mortality. At the hospital level, patients admitted in intensive therapies, immunosuppressed patients, suffering from neutropenia are at high risk of developing invasive candidiasis.

    The scenario of symptoms and clinical signs in cases of candidiasis is broad and varied and may be non-specific. Providing rapid treatment with the correct antifungal is essential. Given the different resistance patterns, mostly species-specific, rapid identification of the species is crucial to choose the most suitable antifungal in case data on antimicrobial susceptibility are not available.

    The Aspergillus genus includes ubiquitous filamentous fungi commonly present in various environments, both outdoors and indoors. Of the over 250 known Aspergillus species, fewer than 40 are of clinical interest. The main route of infection in humans occurs through inhalation of spores produced by the fungus. Allergic bronchopulmonary aspergillosis and chronic pulmonary aspergillosis are the most common infections caused by this pathogen and mainly affect patients with asthma or cystic fibrosis. Invasive aspergillosis, less frequent, is found in immunocompromised patients and can lead to infections with high mortality and morbidity rates. Treatment varies depending on the type of aspergillosis and may include the use of antifungal drugs or the need for surgery.

    The main etiological agent of aspergillosis is Aspergillus fumigatus. However, other Aspergillus species are emerging as pathogens that can also cause severe invasive infections, including A. terreus, which is particularly resistant to antifungals used in aspergillosis therapies, as well as A. flavusA. glaucusA. nidulansA. niger, and A. ustus. A rapid, accurate and reliable diagnosis is essential for an appropriate treatment plan, allowing to reduce the time spent in intensive care units and increase survival rates.

    Human Papillomavirus (HPV) infection is the most common and widespread of sexually transmitted diseases. The different types of HPV are generally divided into low-risk and high-risk cancer risk genotypes. The latter (in particular HPV 16 and 18) are recognized as the necessary cause of cervical cancer and, in general, among the most important carcinogenic viruses for the human species. The transforming role, in fact, does not end in genesis of the cervical carcinoma, but it is strongly correlated to other neoplasms of the female and male genital sphere (vulva, vagina, anus, penis) and extragenital neoplasms (oral cavity, pharynx, larynx). The carcinogenic potentials can be ascribed to two viral oncoproteins (E6 and E7) which, after the integration of the viral genome into the host genome, cause an uncontrolled proliferative stimulus. This leads to a decrease capability in controlling the cellular surveillance mechanisms, with consequent accumulation of genetic abnormalities, increased genomic instability and the appearance of aneuploidy.
    During the process of viral integration, part of the HPV DNA may be lost. However, this process never involves the E6 and E7 genes, so using a screening system targeting those genes exclude the possibility of false negative results due to viral integration events.
