• HRP Substrates (May 02, 2024)

  Soluble Horseradish Peroxidase (HRP) Substrates

  In a soluble chromogenic HRP assay setup, like an ELISA, the preferred substrate is TMB (3, 3', 5, 5'-tetramethylbenzidine). When developing immunoassays, choosing the right TMB is a vital step for optimizing the assay. Kementec offers a range of TMBs with different kinetics and sensitivities, enabling assay developers to choose the right TMB for their assay.

  

  All the TMBs are ECO-TEK, making them REACH, Annex XVII0 compliant and easy to transport with less restrictions. ECO-TEK includes:

  • Free of BSA
  • Free of NMP or other harmful organic solvents – 100% aqueous
  • Free of toxic preservatives and hazardous stop solutions

  The TMBS are characterized by outstanding sensitivity, stability and stop-stability, and very high lot-to-lot consistency. Offering a shelf life up to 4 years of the ready-to-use substrates enables Kementec products to meet all the requirements of assay manufacturers.

  

   

  Several of the TMBs are suitable for use in electrochemical assays that utilize sensor measurements. 

  TMB substrates are also available as unique color-coded products for traceable pipetting. The pink indicator makes manual pipetting traceable, while in automatic analyzers it offers a convenient tracking system for reagent supply. The color disappears after adding the recommended stop solution and does not interfere with the assay.

  Corresponding diluents are available for TMB substrates in order to achieve a specific activity level.

  Over the past decade, immunoassay manufacturers have been working towards more environmentally conscious and sustainable manufacturing. Much of this effort has been focused on reducing usage of facility water and energy use, reducing facility waste, increasing facility recycling, and sourcing recycled/recyclable products packaging.

  Many common components used in ELISAs contain biological and chemical hazards, which were traditionally used to increase assay quality and to prolong assay shelf-life. Considering that the average automated immunoassay analyzer in a clinical lab can run 200 tests per hour while processing samples 24/7, these components can generate a considerable amount of hazardous waste. ELISAs have not traditionally been eco-friendly and several components in this type of assays are usually environmentally harmful.

  Kementec’s environmentally conscious options for immunoassays are called ECO-TEK®.

  ECO-TEK® products are environmentally conscious components enabling development and production of cost efficient immunoassays without compromising quality, stability and accuracy. Additionally, transport of immunoassay may be done worldwide with less import restrictions and paperwork.

• UNI-StabilPLUS (May 02, 2024)

  UNI-StabilPLUS is a universal stabilizer for the dilution and stabilization of both Horseradish Peroxidase (HRP) and Alkaline Phosphatase (AP) labeled proteins and antibodies, in order to maintain the molecular conformation and prevent loss of activity over time. This enables the making of pre-diluted, ready-to-use conjugates, minimizing assay errors in dilution.

  Superior stabilization of HRP and AP conjugated antibodies in low as well as high protein dilutions is seen, when using UNI-StabilPLUS.

  When tested with AP conjugated antibody stability is seen as follows:

  • at least 3 years at 2-8 °C
  • at least 2 years at room temperature
  • at least 4 weeks at 37 °C

  When tested with HRP conjugated antibody stability is seen as follows:

  • at least 2 years at 2-8 °C
  • at least 1 year at room temperature
  • at least 2 weeks at 37 °C

  UNI-StabilPLUS is recommended for the dilution of antibodies directed against rabbit immunoglobulins.

• Enhanced Streptavidin-HRP (May 02, 2024)

  Cat. No. 4740

  Enhanced Streptavidin-HRP is a HRP-conjugate specific for biotinylated antibodies and is characterized by an enhanced molar ratio of >5 HRP enzymes covalently coupled per Streptavidin molecule. Enhanced StreptavidinHRP conjugate enables at least a 4-fold increase in sensitivity in ELISA compared to conventional conjugate and offers pg/fg sensitivity levels in immunoassays, when only minimal amounts of specific antibodies or antigens are available:

  • Signal amplification
  • Ultrasensitive detection
  • Faster assay time
  • Recommended when only minimal amounts of specific antibody or antigen are available
  • Can be used with any biotin labeled probe
